Background
BAFF receptor (BAFF-R/TNFRSF13C, CD268), a member of the tumor necrosis factor receptor superfamily, is a type III transmembrane protein that binds its ligand BAFF as a trimer. Via adaptor proteins it transduces signals essential for B-cell survival, development and the size of the peripheral B-cell pool, thereby tuning both innate and adaptive immunity. Elevated BAFF/BAFF-R levels in human serum or tissues prolong the lifespan of autoreactive B cells and are closely linked to the onset and activity of autoimmune diseases such as systemic lupus erythematosus, making BAFF-R a key diagnostic biomarker and the central target of therapeutics including BAFF or BAFF-R inhibitors.
